A comparative study on Factor ¥·-related antigen(F¥·-RAG) and Ulex europacus I agglutinin (UEA-I) as a vascular endothelial cell marker


Abstract
To compare the usefulness of factor VIII-related antigen (FVIII-RAG) and Ulex europaeus I agglutinin (UEA-I) as a vascular endothelial cell marker, immunoperoxidase staining was performed in the various human normal tissues and vascular neoplasia using peroxidase-antiperoxidase (PAP) and avidinbiotin peroxidase complex (ABC) methods.
The results obtained are as follows:
1. In normal control group, FVIII-RAG showed strong positive staining in endothelial cells of all types of blood vessels of lung, spleen, stomach, cerebrum and endometrial decidua, but focal or weak positive staining in those of kidney, liver, bone marrow, chorionic villi, ovary and salpinx. The endothelium of lymphatic vessel was negative for FVIII-RAG. UEA-I showed strong positive staining in endothelial cells of all normal tissues examined. Lymphatic vessels was positive for UEA-1. UEA-I was also present in the epithelial cells of renal collecting ducts, intrahepatic bile ducts, bronchial mucosa, alveolar lining cells, squamous epithelium of skin, eccrine ducts, gastric mucosa, coelomic epithelium of ovary and salpingeal mucosa.
2. In the vascular neoplastic group, FVIII-RAG showed strong positive staining in endothelial cells of capillary, cavernous and Masson¢¥s hemangiomas, angiomatosis and angiosarcoma, but weak staining in those of pyogenic granuloma and hemangiopericytoma. Lymphangiomas were negative for FVIIIRAG. UEA-I showed strong positive staining in endothelial cells of benign and malignant vascular neoplasia. Lymphangiomas were positive for UEA-I.
3. In the normal and vascular neoplastic groups, the staining of FVIII-RAG was granular in the cytoplasm, whereas that of UEA-I was homogeneous in the cytoplasm and cell membrane.
Above results suggest that FVIII-RAG and UEA-I receptor are reliable markers for the identification of endothelium in normal and tumorous tissue. And it is assumed that UEA-I is more senisitive than anti FVIII-RAG antibody but anti FVIII-RAG antibody is more specific than UEA-I.
